I recently started to prime my 54 235 after rebuild in anticipation of the first start. Unfortunately, as soon as my gauge started to read about 45psi, the Fram bypass filter began to leak out of the top where the wingbolt goes through the lid. I changed the O-ring and even added one so there is an o-ring on the inside and outside of the lid where the wingbolt comes through, but still leaked oil. Has anyone had any experience with this and/or recommend the correct o-ring and location (inside/outside)?

It will seal much better than an O ring. Don't be afraid to tighten it firmly- - - -those things are hard to seal.

Does the standpipe or the cover have a groove for the O ring to fit into?

Mine came with an inverted cup shaped washer at the top with a rubber material in the 'cup' to provide the seal. The rubber had turned hard. I bought a rubber washer at the hardware store to replace the old hard rubber. It seals just fine. I can't say it came new with that setup but it works for me. This is mine.

I put a flat washer and a copper washer on the wingbolt outside the filter, and a fat rubber o-ring on the inside of the lid. No leaks from the filter on the next priming run!!
Unfortunately, the swivel adapters that came with the oil lines BOTH leak, but this seems like a simpler fix - replacements on the way.
