You don't say whether you already have a title or not. If you don't have a title now, they will use your vehicle serial number from the left doorpost, not the engine number.
If you don't have a title it will have to be inspected by the highway patrol since it is not a pre-1950 vehicle.
Make sure you call the highway patrol inspection station before you go to make sure of the hours and whether or not this is a special inspection requiring you to go only during certain hours.
Then I believe, you can take that inspection document, your bill of sale, proof of insurance and your checkbook to a county DMV.

Spend some time on the KS DMV website or call your county DMV office. Their answers to your questions are the only ones that matter.

Kansas was an engine number state through '55. If it's been titled off road or out of state it'll need to be inspected. The inspection consists of a trooper comparing the engine number to the VIN on the title. If you don't have a current title you need to contact your county treasurer (where you get your tags). They can give you all the info you need about what you need to do. My 38 was titled off road so I got to jump through the hoops.
