I would like to use a '52-'54 door latch and striker on my 1949 3600. Will the latch mechanism fit into the door with the same screw points as the '49. Can I put it in "un-modified"?

Nope. And the other issue you have is that the door handles are different and you don't have the backing plate on the inside of the door to run the screw into the handle. I have a '51 and a '53. Door handles were changed in 52. I wanted to put the '53 style handles on my '51 and decided it was too hard.

I wont to put in my chevy 3600 47-51 door latch and striker in my 1952 chevy,opposite than you, the latchs holes are not the same, the 49 ext handle not fit into the 1952 door, the 1949 drive door has the hole for the lock, the 1952 not because this lock is the same push botton.
In summary, don't work without any modification
