Both of these engines have oil gallies which are plugged on the outside of engine block (front and rear.) I’ve read reports that these plugs can leak or dislodge. There are recommendations to tap these ports and plug with pipe plugs.
Is it recommended for the 216 and 235?

Yes, also small block V8 engines. I use Allen socket head pipe plugs- - - - -1/4" NPT. Be sure to clean out all the cuttings caused by the threading operation!

I've used Permatex Super 300 brush-on liquid for many years, but it's getting very hard to find these days. Any PTFE (Teflon) brush-on pipe sealer is a good substitute when installing pipe thread fittings, but it can't be used as a general purpose gasket sealer like Super 300. NAPA still lists the stuff online, but I've had problems actually ordering it at the local NAPA store. The chocolate brown color Permatex "3-D" and "3-H" brush on sealer is not the same as Super 300, which was originally developed for use around aviation gasoline. Super300 is almost black, and does a much better job of sealing and staying pliable under heat and pressure. Putting brass pipe plugs into cast iron usually seals well without any liquid help.

Super 300 has been discontinued according to Permatex. I purchased a bottle about 2 years ago on Amazon to do Ol' Roy's water pump, I used it recently for the pushrod cover and valve cover.

Permatex recommends a substitute for Super 300 called Aviation Form-A-Gasket #3.

Part #80017 or #80019 (part numbers refer to different size bottles)

Permatex #3 resembles a vacuum cleaner- - - -it "inhales vigorously!" Those two different size bottles used to be identified as "3D" and "3H".

Pulling those gallery plugs is also an important part of a good rebuild- - - - - -it allows the oiling system passages to be scrubbed out with a bronze shotgun cleaning brush and solvent. It's surprising how much crud can accumulate in the oil system passages over a few decades, and how much damage it can do to new bearings. Especially after some cleaning has been done during rebuild prep- - - - -loosening up that crud so it can wreak havoc on that fresh rebuild!
