I have a door that someone has stripped the threads that attach the hinge to the door. Has anyone tackled this problem before? Can the plates be removed from the door to fix or fab new ones?

If this were my issue, it wouldn't bother me one bit to over drill the holes and tap to the next size up.

Another option besides using larger bolts (which may reduce door alignment adjustment ability) is to install helicoils in the stripped holes. The original bolt size is 3/8-24 and the next readily available size is 1/2-20, (7/16-20 bolts are available, but a little harder to find) so you'd loose 1/8" overall movement without hogging out the slots in the hinge bars. Helicoil kits are pretty easy to use, and come with the proper tap, inserts and an installation tool.

You could theoretically remove and replace the nut plate from the door, but access is really limited. I wouldn't want to try that. Too many opportunities to get it wrong or mess something else up.

Depends on what year truck, if both doors are stripped and which holes are stripped, top or bottom and how many. I can offer ideas if I know what truck and etc. I had to search WAY back to get a possible 1952 big bolt post mention.

Several good ideas have been given already.

Are the bolt threads stripped or are the captive nuts stripped? The captive nuts are usually made of harder steel than the bolts.
Have you tried a new bolt to check the condition of the nuts?

I'm not sure the exact year as I bought them they have vent windows and push button handles so 52 up maybe?

Update; Drilled and taped to 7/16"-20 all is well.
