I am having issues with the DS door on my 56 3100. It literally jumps up and down as i pick up speed and am on uneven highway. My door does not have weather strip in it as of now, is that likely the main issue causing this? Or could it be an alignment issue? The door closes and opens good, looks aligned from the outside. I have the weather-strip, but am holding off putting it in until i paint the inside of the cab. I am basically doing a build, area by area, so i can drive and enjoy it during the process. Hope that makes sense. Appreciate any tips or info around this.

Without the weather strip it is going to “bounce” around. It’s really all the flexing of the cab and it not having any cushion
Unless the latch is not holding. Mine is a 56 and that’s what it seemed like till I replaced the weather strip.

The "jumps up and down" make me want to suggest you check your hinges while your at it. Open the door wide open and grab the lower part of the door and lift up on it and check to see how much "up and down" play you have. I suspect you will find you have "a lot". Rebuild the hinges if that's what you find.

I checked my drivers door and it is solid from moving up and down - no play that way at all, but it has like a half inch of play moving out and back in again. I was incorrect in saying it had up and down motion in my first post. It just seemed that way when i was driving it on a rough road. I think adding the weather stripping will help. But curious if anyone else has had this issue of the door moving out and in, and if rebuilding the hinges would still help with this? Thanks.

Unless it moves in and out at the hinge pillar, then rebuilding the hinges won't help. The weatherstrip is likely your main issue, and after that's installed, adjusting the latch striker should take care of it.
