Yes, you will need to lock the hubs first. You can leave the hubs locked and shift from 2 to 4 wheel drive "on the fly" as long as you keep it under 25mph. Low range will require stopping. Leaving the hubs locked when you think you'll need to go into or out of 4wd will not hurt anything, the frond drive components will just spin freely when in 2wd mode. You do want to unlock the hubs when 2wd is going to be used for long periods to decrease wear and improve fuel efficiency.
