If this all needs to come apart and out, will I need to take the entire assembly out, axles brakes etc, all of it? Or do I take apart just the pumpkin?
Should I have a real transmission shop set-up the gears? or is this a DIY job?

The axle shafts will need to be removed before you can remove the Pumpkin from the rear end.
You could do a DIY rebuild on your rear end. I've done quite a few different US and European rear end rebuilds now, and they've all worked so far -there's no rocket science involved in a rebuild, but the ring, pinion and differential do need to be assembled with exactly the right settings to run reliably and quietly. If you've never done a rear end rebuild before, you'd do best to read a good manual before you think about taking anything apart. This .pdf document isn't specific to any particular rear end rebuild but will give you good idea of what's involved to do the job properly.
