The lower fitting toward the front of the block should be the supply line TO the full flow filter. Output from the filter needs to be plumbed to the upper, rearward fitting. Use at least 3/8" diameter braided pressure hose- - - -1/2" is better. If the filter adapter is mounted to the body or frame, use flex hoses, not hard lines. You can hard-plumb the filter if it's mounted to the block or intake manifold. The small 1/8" pipe port beside the front fitting can be plugged- - - -it was originally the return port from a bypass filter and all it does is drain back into the oil pan.
