My question is this, I am thinking of going to the closed drive line. If I change the rear end, will I lose that sound?

What transmission and rear end do you have?
What transmission will you change-to?
An original 1952 Chevrolet 1/2 ton truck would have had a closed drive line (torque-tube, enclosed drive-shaft) and a 4.11 ratio rear end.
