I’ve never done this, because I never had any rubber between pump and carb. But I rigged up a temp setup to test out my “new” YF…so figure I might as well check fuel pressure.
My vac gauge reads vac and pressure, supposedly. Just tee it into the line and that’s all there is to it?

That's one third of a fuel pump test. Drop me a PM for the complete procedure. No, a T fitting in the line won't work properly. Jerry

While we're at it, what is the specified amount of fuel that should be pumped in a specified number of squirts?
My Cadillac requires 8 oz of fuel to be pumped into a jar with 9 squirts of the pump. It's very simple and straight forward.
Does the 216 and its brothers have a similar requirement that would be easy to test?

I've always used the rule of thumb "pumps a pint of fuel or more into a catch container at idle speed in 30 seconds". If the engine won't idle for half a minute on the gas in the float bowl, it has other problems. I've got my steel pot and flak jacket on, so let's go ahead and cue up the "Don't have an open fuel line on a running engine" crowd- - - -I know they're just waiting to pile on! LOL! Jerry

I heard “open fuel line on running engine” is what killed Meatloaf.

I was taught as a young lad, "not to play with fire." I am no longer a young lad. Neither was Red Adair. He played with fire. Nobody ridiculed him. I still want a tin foil fire suit and hat.
