Grease the suspension

Is the best way to grease fittings (spring shackles, king pins, drag link, etc) by putting the truck on jack stands to take weight off the joint? Or does it matter?

Kingpins yes. They are tight. The rest are petty loose and grease passes through pretty easily.

...and if you happen to have a helper around it sometimes will be nice to have them push up and down on the front or read as well as have them turn the wheels on the front with it jacked up. "A little bit of sugar helps the medicine go down".

I like to jack up the end I am working on to relieve the pressure, grease the fittings, then I lower it and grease the fittings again.
I also love my Dewalt power grease gun..
